Effects of Lactobacillus curvatus HY7601 and Lactobacillus plantarum KY1032 on Overweight and the Gut Microbiota in Humans: Randomized, Double-Blinded, Placebo-Controlled Clinical Trial

Methods
Randomized, double-blind, placebo-controlled clinical trial in overweight/obese Korean adults. Participants: 72 adults aged 19–65 years with BMI 23.0–<35 kg/m2; both genders; generally healthy; conducted at Vievis Namuh Hospital, Seoul, Korea. 30 assigned to probiotics and 29 to placebo.
Intervention
One 350 mg capsule taken daily after breakfast for 12 weeks, containing 250 mg Lactobacillus curvatus HY7601 and 250 mg Lactobacillus plantarum KY1032 (5×10^9 CFU each per capsule).
Results
Over 12 weeks, HY7601 and KY1032 supplementation yielded modest weight loss and improvements in central adiposity and adipokines versus placebo. Weight change: probiotic −0.47 kg vs placebo +0.93 kg (between-group difference significant, p=0.001); BMI −0.15 kg/m2 vs +0.32 kg/m2 (p<0.001); waist circumference −0.41 cm vs +1.31 cm (p=0.007); body fat mass −0.28 kg vs +0.28 kg (p=0.043); visceral fat area −1.67 cm2 vs +2.82 cm2 (p=0.025); lean mass −0.18 kg vs +0.51 kg (p=0.032). Adiponectin and leptin changes differed between groups (adiponectin p=0.046; leptin p=0.03). Probiotics increased beneficial gut taxa (Bifidobacteriaceae, Akkermansiaceae) and decreased obesity-associated taxa (Prevotellaceae, Selenomonadaceae); species-level increases included Bifidobacterium adolescentis, B. longum, and Akkermansia muciniphila, with decreases in Collinsella aerofaciens, Faecalibacterium prausnitzii, and Prevotella copri. Correlations showed B. adolescentis and A. muciniphila linked to lower obesity measures; P. copri and Megamonas funiformis linked to higher measures. Conclusion: HY7601 and KY1032 can modulate the gut microbiome and body weight in overweight adults, supporting potential use as a dietary intervention to prevent or alleviate obesity.
Limitations
Small, single-country sample with dropouts (64 completers from 72 enrolled); short duration (12 weeks); modest effect sizes; results may not generalize to other populations or individuals with comorbidities.
